=== Intragenic complementation === When multiple copies of a polypeptide encoded by a gene form a complex, this protein structure is referred to as a multimer. When a multimer is formed from polypeptides produced by two different mutant alleles of a particular gene, the mixed multimer may exhibit greater functional activity than the unmixed multimers formed by each of the mutants alone. In such a case, the phenomenon is referred to as intragenic complementation (also called inter-allelic complementation). Intragenic complementation has been demonstrated in many different genes in a variety of organisms including the fungi Neurospora crassa, Saccharomyces cerevisiae and Schizosaccharomyces pombe; the bacterium Salmonella typhimurium; the virus bacteriophage T4, an RNA virus and humans. In such studies, numerous mutations defective in the same gene were often isolated and mapped in a linear order on the basis of recombination frequencies to form a genetic map of the gene. Separately, the mutants were tested in pairwise combinations to measure complementation. An analysis of the results from such studies led to the conclusion that intragenic complementation, in general, arises from the interaction of differently defective polypeptide monomers to form a multimer. Genes that encode multimer-forming polypeptides appear to be common.
== Extramitochondrial localisation == Cytochrome c is widely believed to be localised solely in the mitochondrial intermembrane space under normal physiological conditions. The release of cytochrome c from mitochondria to the cytosol, where it activates the caspase family of proteases, is believed to be the primary trigger leading to the onset of apoptosis. Measuring the amount of cytochrome c leaking from mitochondria to cytosol, and out of the cell to culture medium, is a sensitive method to monitor the degree of apoptosis. However, detailed immuno-electronmicroscopic studies with rat tissues sections employing cytochrome c specific antibodies provide compelling evidence that cytochrome c under normal cellular conditions is also present at extramitochondrial locations. In pancreatic acinar cells and the anterior pituitary, strong and specific presence of cytochrome c was detected in zymogen granules and in growth hormone granules, respectively. In the pancreas, cytochrome c was also found in condensing vacuoles and in the acinar lumen. The extramitochondrial localisation of cytochrome c was shown to be specific as it was completely abolished upon adsorption of the primary antibody with purified cytochrome c. Besides cytochrome c, extramitochondrial localisation has also been observed for large numbers of other proteins including those encoded by mitochondrial DNA. This raises the possibility of the existence of yet-unidentified specific mechanisms for protein translocation from mitochondria to other cellular destinations.
== Access == The library is open for academic research to anyone with a Reader Identification Card. One may not remove library items from the reading rooms or the library buildings. === Inducers === Many compounds commonly used in ferroptosis studies including erastin, RSL3 (RAS-selective lethal), ML162, and ML210 [from National Institutes of Health-Molecular Libraries Small Molecule Repository (NIH-MLSMR)] were initially identified in screens for compounds that can selectively kill cancerous mutant RAS cells. Initial studies characterized the mitochondrial VDAC2 and VDAC3 as the targets of erastin, though it was later found that the mechanistic target of erastin is the cystine/glutamate transporter system xc−. Erastin inhibits system xc−, lowering intracellular GSH levels. Consequently, the GSH-dependent GPX4 is unable to detoxify lipid hydroperoxide species, leading to ferroptotic cell death. Derivatives of erastin have been prepared to improve aqueous solubility, potency, and metabolic stability, with imidazole ketone erastin (IKE) being the most extensively studied. RSL3 and ML162 contain chloroacetamide moieties that can covalently react with nucleophilic residues. RSL3 and ML162 are able to bind to and inhibit GPX4 enzymatic activity or degrade GPX4 in lysate-based assays, though it has been found that RSL3 and ML162 do not inhibit purified GPX4 in vitro and target other selenoproteins such as thioredoxin reductase 1 (TXNRD1). However, other TXNRD1 inhibitors do not trigger ferroptosis, suggesting that TXNRD1 inhibition is not sufficient to trigger ferroptosis. The GPX4-inhibiting activity of RSL3 has also been suggested to be regulated by other factors such as 14-3-3ε or through broad targeting of the selenoproteome.

Pancreatic ductal cells are the epithelial cells that line the pancreatic ducts which deliver enzymes from the acinar cells to the duodenum. They have the essential function of producing bicarbonate-rich (HCO3-) secretion to neutralize stomach acidity. The hormone secretin stimulates ductal cells and is responsible for maintaining the duodenal pH and preventing duodenal injury from acidic chyme. Ductal cells mix their production with acinar cells to make up the pancreatic juice. Ductal cells comprise about 10% of the pancreas by number and about 4% in volume. Its function is to secrete bicarbonate and mucins and to form the tubule network that transfers enzymes made by acinar cells to the duodenum. Ductal cells have a proliferation rate of about 0.5% in normal adults, but mitotic activity goes up when the pancreas is damaged.
Descended from Howard Fisher of the Fisher Brothers, owners of Fisher Body, from 1919 a part of General Motors, she has served as president of the General Motors Foundation and as executive director of global community relations and government relations at GM. Their marriage lasted 38 years until her husband's death on February 7, 2019, at the age of 92. Like her husband, she is a Catholic. She is a member of the Democratic National Committee from Michigan and chaired Vice President Al Gore's campaign in Michigan in 2000. In 2004, she also helped secure the Michigan Democratic primary and general election vote for John Kerry in Michigan. In November 2006, Dingell was elected to Wayne State University's board of governors. Dingell and Senator Carl Levin were proponents of moving up Michigan's presidential primary before February 5 in an attempt to garner greater political influence for Michigan during the 2008 Democratic primaries. This resulted in Michigan almost losing its delegates' votes in the Democratic National Convention. When Carl Levin announced his retirement from the U.S. Senate at the end of his term in 2015, Dingell indicated that she was interested in running for his seat. When former Michigan Governor Jennifer Granholm declined to run for the seat, a Politico writer declared Dingell to be one of the front-runners for the Democratic nomination, alongside Representative Gary Peters. She chose not to run, and Peters won the seat.

Photosynthesis is the primary process by which carbon moves from the atmosphere into living things. In photosynthetic pathways 12C is absorbed slightly more easily than 13C, which in turn is more easily absorbed than 14C. The differential uptake of the three carbon isotopes leads to 13C/12C and 14C/12C ratios in plants that differ from the ratios in the atmosphere. This effect is known as isotopic fractionation. To determine the degree of fractionation that takes place in a given plant, the amounts of both 12C and 13C isotopes are measured, and the resulting 13C/12C ratio is then compared to a standard ratio known as PDB. The 13C/12C ratio is used instead of 14C/12C because the former is much easier to measure, and the latter can be easily derived: the depletion of 13C relative to 12C is proportional to the difference in the atomic masses of the two isotopes, so the depletion for 14C is twice the depletion of 13C. Some studies suggests that inadequate published data in animal testing may result in irreproducible research, with missing details about how experiments are done are omitted from published papers or differences in testing that may introduce bias. Examples of hidden bias include a 2014 study from McGill University which suggests that mice handled by men rather than women showed higher stress levels.
