Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.
The compound was developed during the 1980s at the Institute of Molecular Genetics in Moscow as part of research on fragments of adrenocorticotropic hormone. Early work examined short ACTH-derived sequences that retained neurotrophic effects while lacking the endocrine activity of the full hormone. Semax entered clinical use in Russia during the 1990s, where it received registration for several neurological indications. Outside that region it remained primarily a laboratory research material rather than an approved therapeutic. English-language literature on it grew more slowly and frequently cited the original Russian studies.
Published studies examine a fairly narrow set of endpoints. Rodent experiments commonly measure maze learning, infarct volume after induced ischemia, and tissue levels of neurotrophic factors. Clinical reports from Russian centres describe attention, memory and recovery scores in patients after stroke or transient ischemic attack. Most of those human studies are small and few have been repeated by independent groups. Outcome measures differ between studies, which limits direct comparison.
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.

Scientific literature on semax is unevenly distributed. A substantial share of published work originates from a small number of laboratories in Russia, while independent replication elsewhere is limited. Human data consist mostly of small trials with short follow-up, and several reported outcomes rely on subjective rating scales. Questions about how much intact peptide reaches the central nervous system after nasal administration, and how long it persists there, are still unresolved. The compound is best described as an active research subject rather than a settled pharmacological agent.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It was derived from the ACTH(4-10) fragment, a short segment of adrenocorticotropic hormone that lacks the hormonal activity associated with the full-length peptide. Researchers at the Institute of Molecular Genetics in Moscow developed the compound during the 1980s. It has been registered as a pharmaceutical product in Russia and several neighbouring countries, where it is supplied as a nasal solution, and it is also sold internationally as a research chemical.
The proposed mechanism centres on neurotrophic signalling rather than direct receptor activation. Semax is reported to increase expression of brain-derived neurotrophic factor and nerve growth factor in several brain regions, and to shift the balance between excitatory and inhibitory neurotransmitter systems. Interaction with melanocortin receptors has been suggested because of the parent ACTH fragment. Many of these findings come from rodent studies, and the extent to which they translate to human physiology remains an open question.
== Signal peptide as a therapeutic target == Signal peptide is a potential (therapeutic) antiviral target. The stable signal peptides (SSPs) of mammarenavirus viral glycoprotein precursors re-associate with mature spike proteins after their co-translational cleavage. SSPs with a penultimate N-terminus glycine are a target for NMT inhibitors, which inhibit the myristoylation of signal peptides and target the signal peptide for proteasomal degradation, affecting virus–cellular fusion.
== Adverse effects == In a systematic review analyzing data from 5 cohort studies having 1,085,488 patients, use of gabapentinoids (pregabalin and gabapentin) was associated with an increased risks of thrombotic events (deep venous thrombosis and pulmonary thrombo-embolism) as early as three months of use, and with increased risk of cardiovascular events on prolonged use of more than a year duration. Heart failure was not increased with the use of gabapentinoids. Exposure to pregabalin is associated with weight gain, drowsiness, fatigue, dizziness, vertigo, leg swelling, disturbed vision, loss of coordination, and euphoria. It has an adverse effect profile similar to other central nervous system (CNS) depressants. Even though pregabalin is a depressant and anticonvulsant, it can sometimes paradoxically induce seizures, particularly in large overdoses. Adverse drug reactions associated with the use of pregabalin include:
==== Canada ==== As of 11 April 2020, access in Canada was available only through clinical trials. Health Canada approved requests to treat twelve people with remdesivir under the department's special-access program (SAP). Additional doses of remdesivir are not available through the SAP except for pregnant women or children with confirmed COVID‑19 and severe illness. In June 2020, Health Canada received an application from Gilead for the use of remdesivir for treating COVID‑19. On 27 July 2020, Health Canada conditionally approved the application. In September 2020, Minister of Public Services and Procurement Anita Anand announced that Canada had entered into a deal to obtain up to 150,000 vials of remdesivir from Gilead starting in October. As of 8 October, remdesivir was still not widely available in Alberta, because Alberta Health Services was undertaking a "formulary review" to be completed by mid-November.

Barrel-stave model: The barrel-stave model proposes that AMPs interact with the lipid bilayer of the microbial cell membrane to form transmembrane channels or "barrel staves". These channels are thought to disrupt the membrane's integrity, leading to the death of the microbe. Carpet model: The carpet model proposes that AMPs adsorb onto the lipid bilayer of the microbial cell membrane, forming a dense layer that causes the membrane to become permeabilized. This model suggests that the AMP acts as a "carpet" that covers the surface of the cell, preventing the microbe from functioning properly. Toroidal model: The toroidal model proposes that AMPs interact with the lipid bilayer of the microbial cell membrane to form toroidal structures, which are thought to pinch off sections of the membrane and lead to the formation of vesicles. This process is thought to disrupt the membrane's integrity and cause the death of the microbe. Disordered toroidal-pore model: According to this model, the disordered AMPs wrap around the lipid bilayer and create a pore, which disrupts the membrane's integrity and leads to the death of the microbe. Unlike the toroidal model, which suggests that the AMP creates a stable toroidal structure, the disordered toroidal-pore model suggests that the AMP is flexible and does not form a stable toroidal structure. The peptide-lipid pore complex becomes intrinsically disordered, with the orientation of the peptide not well defined.
Hydrocodone/paracetamol (also known as hydrocodone/acetaminophen) is the combination of the pain medications hydrocodone (an opioid) and paracetamol (acetaminophen). It is used to treat moderate to severe pain. It is taken by mouth. Recreational use is common in the United States. Common side effects include dizziness, sleepiness, constipation, and vomiting. Serious side effects include addiction, decreased rate of breathing, low blood pressure, severe allergic reactions, and liver failure. Use during pregnancy may harm the fetus. Use with alcohol is not recommended. Hydrocodone works by binding to the mu-opioid receptor. How paracetamol works is unclear but may involve blocking the creation of prostaglandins. Hydrocodone/paracetamol was approved for medical use in the United States in 1982. In the United States, it is a schedule II controlled substance. In 2023, it was the 25th most commonly prescribed medication in the United States, with more than 21 million prescriptions. It is not available in the United Kingdom, though the combination codeine/paracetamol (co-codamol) is. It is sold under the brand names Vicodin and Norco among others.
He conceived an idea that if a molecule that could block the enzyme is discovered, it could become an immunosuppressive drug that could be used for autoimmune diseases and in organ transplantation. In 1981 he decided to go for drug discovery and approached several pharmaceutical companies, which turned him down one by one as he had no primary knowledge of drug research. However, Syntex liked his plans and asked him to join the company with his wife. He became vice president for the research. In one of their experiments the Allisons used an antibacterial compound, mycophenolate mofetil, which was abandoned in clinical use due to its adverse effects. They discovered that the compound had immunosuppressive activity. They synthesised a chemical variant for increased activity and reduced adverse effects. They subsequently demonstrated that it was useful in organ transplantation in experimental rats. After successful clinical trials, the compound was approved for use in kidney transplant by the U.S. Food and Drug Administration on 3 May 1995, and was sold under the brand name Cellcept. It was approved for use in the European Union in February 1996.

...some of the actions adopted by those aiming to stop tours were despicable. Bette [Bill McLaren's wife] and I were referred to as 'racist scum' on our way down to Mansfield Park to see the South African Barbarians play in 1979 – a tour party comprising equal numbers of coloureds, blacks and whites. That, said the protesters, was just window dressing. When the Springboks last toured the UK, I was asked to provide commentary on their match against the Midland Counties (East) at Welford Road, Leicester. I remember having to walk the gauntlet up a narrow channel lined on each side by policemen holding back the mob. Those policemen were covered in spittle, had hats knocked off, were kicked in places where no man should be kicked, and yet took it all with stoic calm. I couldn't believe that people in the British Isles would behaved in that manner.... Constant noise outside the South Africans' hotels to try and prevent them sleeping was another unbelievable ploy that sickened decent people.

Margaret Ruth Kidder (October 17, 1948 – May 13, 2018) was a Canadian-American actress and activist. She amassed several film and television credits in her career spanning five decades, including her widely known role as Lois Lane in the original Superman films (1978–1987). Her accolades included two Canadian Film Awards, an Emmy Award, a Genie Award, and a Saturn Award. Born in Yellowknife to a Canadian mother and an American father, Kidder was raised in the Northwest Territories and several Canadian provinces. She began her acting career in the 1960s, appearing in low-budget Canadian productions and winning the Canadian Film Special Award in 1969. She first received attention for appearing in the comedy film Quackser Fortune Has a Cousin in the Bronx (1970), the horror films Sisters (1972), Black Christmas (1974), and The Reincarnation of Peter Proud (1975), and the drama films A Quiet Day in Belfast (1974) and The Great Waldo Pepper (1975). Kidder's international breakthrough came with playing Lois Lane in Superman (1978) and Kathy Lutz in The Amityville Horror (1979), which were blockbuster films. For these roles, she was twice nominated for the Saturn Award for Best Actress, winning in 1978 for Superman. She reprised the role of Lois in three Superman sequels (1980–1987), and also played Rita Harris in the comedy film Heartaches (1981) and made her stage debut with the play Bus Stop (1982).
==== Half-Life 2: Lost Coast (2005) and Team Fortress 2 (2007) ==== Antonov worked on Half-Life 2: Lost Coast, an additional level for Half-Life 2 released in 2005 as a free download for owners of the game. His last credit at Valve was for the online multiplayer FPS Team Fortress 2 (2007), temporarily working as its art director until his friend Moby Francke joined the company, at which point Antonov handed the role to him. Antonov left the company in 2006. He later explained that he was disappointed he had "spent six years of my life on one single project", and he did not care to work on small-scale episodic games, which Valve made with Half-Life 2: Episode One (2006) and Episode Two (2007). He wanted to make more "AAA" games like the original Half-Life 2, at a job in Europe which gave more artistic independence and "maneuverability" than Valve.
=== Magnetic bead method === New extraction techniques have been developed using magnetic beads for the purification of nucleic acids by taking advantage of the charged and polymeric nature of long strand of DNA. Beads are both uncoated to increase surface are and yield, while others are more selective by being coated with functional groups that interact with the polymers present in microbes. One common method is to use polyethylene glycol to drive DNA binding to the magnetic beads. The molecular weight and concentration of the PEG will control what molecular weight DNA binds.
Gluconeogenesis (GNG) is a metabolic pathway that results in the biosynthesis of glucose from certain non-carbohydrate carbon substrates. It is a ubiquitous process, present in plants, animals, fungi, bacteria, and other microorganisms. In vertebrates, gluconeogenesis occurs mainly in the liver and, to a lesser extent, in the cortex of the kidneys. It is one of two primary mechanisms – the other being degradation of glycogen (glycogenolysis) – used by humans and many other animals to maintain blood sugar levels, avoiding low levels (hypoglycemia). In ruminants, because dietary carbohydrates tend to be metabolized by rumen organisms, gluconeogenesis occurs regardless of fasting, low-carbohydrate diets, exercise, etc. In many other animals, the process occurs during periods of fasting, starvation, low-carbohydrate diets, or intense exercise. In humans, substrates for gluconeogenesis may come from any non-carbohydrate sources that can be converted to pyruvate or intermediates of glycolysis (see figure). From the breakdown of proteins, these substrates include glucogenic amino acids (although not ketogenic amino acids); from breakdown of lipids (such as triglycerides), they include glycerol, odd-chain fatty acids (although not even-chain fatty acids, see below); and from other parts of metabolism that includes lactate from the Cori cycle. Under conditions of prolonged fasting, acetone derived from ketone bodies could potentially also serve as a substrate based on computer models.

It is a synthetic seven-amino-acid peptide built from the ACTH(4-7) sequence plus a Pro-Gly-Pro tail. It belongs to the family of short ACTH fragments studied for nervous-system effects. Its backbone contains only standard amino acids.
It contains the 4-7 fragment of adrenocorticotropic hormone extended at the C terminus. That truncation removes the region associated with steroidogenic activity. The design intent was to keep neurotrophic properties while avoiding full hormonal effects.
No natural source of the intact heptapeptide is known. It is produced by solid-phase peptide synthesis. Some related ACTH fragments arise as hormone processing products, but the Pro-Gly-Pro extension is a designed addition rather than a natural sequence.
It consists mainly of animal experiments and small clinical reports, with much of the clinical material published in Russian-language journals. Large independent trials are scarce. Separating reliable effects from chance findings is consequently difficult.
