The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Solid semax is typically supplied as a lyophilised powder that is hygroscopic and sensitive to moisture, light, and repeated temperature cycling. Long-term storage of the dry peptide is generally recommended at approximately -20 degrees Celsius, while shorter working periods may use refrigeration at 2 to 8 degrees Celsius. Vials should remain tightly closed and desiccated when brought to room temperature, because condensation can damage the material before it is weighed. Dividing a batch into aliquots is preferable to thawing one container repeatedly.
Dissolution is normally performed in water, phosphate-buffered saline, or normal saline, since the peptide is freely soluble in aqueous media and is rarely handled with strong organic solvents. Solution pH should be kept near neutral, because extreme acidity or alkalinity accelerates backbone hydrolysis and encourages oxidation of the methionine side chain. Once dissolved, the material is less stable than the dry powder and is commonly divided into single-use portions and frozen. Buffers containing primary amines, such as Tris, are often avoided because of possible side reactions.
Purity assessment relies mainly on reversed-phase high-performance liquid chromatography, which separates the target heptapeptide from truncated sequences, deletion analogues, and oxidised forms. Mass spectrometry, usually coupled to liquid chromatography, confirms identity through the expected molecular ion and reveals modifications such as methionine sulfoxide formation. Amino acid analysis can verify composition, and tandem mass spectrometry supports sequence confirmation. Ultraviolet detection near 254 to 280 nanometres is convenient because the phenylalanine and histidine residues absorb in that region. Nuclear magnetic resonance is rarely used for routine release testing.

Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
"It really got to them," recalls Dr. William Schneider Jr., [former] undersecretary of state for military assistance and technology, who saw classified "after-action reports" that indicated U.S. flight activity. "They didn't know what it all meant. A squadron would fly straight at Soviet airspace, and other radars would light up and units would go on alert. Then at the last minute the squadron would peel off and return home." From the accounts of CIA and senior KGB officers, by May 1981, obsessed with historical parallels with the 1941 German invasion and Reaganite rhetoric, and with no defensive capability against the Pershing IIs, Soviet leaders believed the United States was preparing a secret nuclear attack on the USSR and initiated Operation RYaN. Under this, agents abroad monitored service and technical personnel who would implement a nuclear attack so as to be able either to preempt it or have mutually assured destruction. On 1 September 1983, the Soviet military shot down a South Korean passenger jet, Korean Air Lines Flight 007, that had strayed into Soviet airspace. All 269 people aboard the aircraft were killed, including U.S. Representative Larry McDonald and many other Americans. The first Pershing II missiles were delivered to West Germany on 1 December 1983. Bruce G. Blair, an expert on Cold War nuclear strategies and former president of the World Security Institute in Washington, D.C., says the American–Soviet relationship at that time:
=== Location === Ghrelin cells are found mainly in the stomach and duodenum, but also in the jejunum, lungs, pancreatic islets, gonads, adrenal cortex, placenta, and kidney. It has also been shown that ghrelin is produced locally in the brain. Additionally, research suggests that ghrelin may be produced in the myocardium and have an 'autocrine/ paracrine' like effect within the heart. Ghrelin cells are also found in oxyntic glands (20% of cells), pyloric glands, and small intestine.
=== Enlarged prostate === Physicians sometimes prescribe finasteride for the treatment of benign prostatic hyperplasia, informally known as an enlarged prostate. Finasteride may improve the symptoms associated with BPH such as difficulty urinating, getting up during the night to urinate, hesitation at the start and end of urination, and decreased urinary flow. The use of the drug showed significant sexual adverse effects such as erectile dysfunction and less sexual desire, in particular when obstructive symptoms due to an enlarged prostate were present.
In 1952, retired chicken incubator salesman George W. Church Sr. opened the first Church's Chicken, named Church's Fried Chicken To-Go, in San Antonio, Texas. Initially, Church's Chicken was a single walk-up establishment that sold only fried chicken. Two pieces of chicken and a roll cost 49 cents. Church's Chicken added fries and jalapeños to its menu in 1955. To allow customers to see their food prepared while they waited, Church Sr. designed the kitchen with the fryers next to the takeout window.
From southern Scandinavia and northern Germany, the Germanic peoples expanded south, east, and west, coming into contact with the Celtic, Iranian, Baltic, and Slavic tribes. Southern Germany was inhabited by Celtic-speaking peoples, who belonged to the wider La Tène culture. They were later assimilated by the Germanic conquerors. Under Augustus, the Roman Empire began to invade lands inhabited by the Germanic tribes, creating a short-lived Roman province of Germania between the Rhine and Elbe rivers. In 9 AD, three Roman legions were defeated by Arminius in the Battle of the Teutoburg Forest. The outcome of this battle dissuaded the Romans from their ambition of conquering Germania and is thus considered one of the most significant events in European history. By 100 AD, when Tacitus wrote Germania, Germanic tribes had settled along the Rhine and the Danube (the Limes Germanicus), occupying most of modern Germany. However, Baden-Württemberg, southern Bavaria, southern Hesse and the western Rhineland had been incorporated into Roman provinces. Around 260, Germanic peoples broke into Roman-controlled lands. After the invasion of the Huns in 375, and with the decline of Rome from 395, Germanic tribes moved farther southwest: the Franks established the Frankish Kingdom and pushed east to subjugate Saxony and Bavaria. Areas of modern eastern Germany were inhabited by Western Slavic tribes.

=== Chronic cholecystitis === Chronic cholecystitis occurs after repeated episodes of acute cholecystitis and is almost always due to gallstones. Chronic cholecystitis may be asymptomatic, may present as a more severe case of acute cholecystitis, or may lead to a number of complications such as gangrene, perforation, or fistula formation. Xanthogranulomatous cholecystitis (XGC) is a rare form of chronic cholecystitis which mimics gallbladder cancer although it is not cancerous. It was first reported in the medical literature in 1976 by McCoy and colleagues.
Thirty years later, this work inspired, independently, two research groups to adopt the H-phosphonate chemistry to the solid-phase synthesis using nucleoside H-phosphonate monoesters 7 as building blocks and pivaloyl chloride, 2,4,6-triisopropylbenzenesulfonyl chloride (TPS-Cl), and other compounds as activators. The practical implementation of H-phosphonate method resulted in a very short and simple synthetic cycle consisting of only two steps, detritylation and coupling (Scheme 2). Oxidation of internucleosidic H-phosphonate diester linkages in 8 to phosphodiester linkages in 9 with a solution of iodine in aqueous pyridine is carried out at the end of the chain assembly rather than as a step in the synthetic cycle. If desired, the oxidation may be carried out under anhydrous conditions. Alternatively, 8 can be converted to phosphorothioate 10 or phosphoroselenoate 11 (X = Se), or oxidized by CCl4 in the presence of primary or secondary amines to phosphoramidate analogs 12. The method is very convenient in that various types of phosphate modifications (phosphate/phosphorothioate/phosphoramidate) may be introduced to the same oligonucleotide for modulation of its properties. Most often, H-phosphonate building blocks are protected at the 5'-hydroxy group and at the amino group of nucleic bases A, C, and G in the same manner as phosphoramidite building blocks (see below). However, protection at the amino group is not mandatory.
== Epidemiology == A direct inguinal hernia is less common (~25–30% of inguinal hernias) and usually occurs in men over 40 years of age. Men have an 8 times higher incidence of inguinal hernia than women.
The inhibitory effect of the available GABA is potentiated, leading to sedative and anxiolytic effects. For instance, those ligands with high activity at the α1 are associated with stronger hypnotic effects, whereas those with higher affinity for GABAA receptors containing α2 and/or α3 subunits have good anti-anxiety activity. GABAA receptors participate in the regulation of synaptic pruning by prompting microglial spine engulfment. Benzodiazepines have been shown to upregulate microglial spine engulfment and prompt overzealous eradication of synaptic connections. This mechanism may help explain the increased risk of dementia associated with long-term benzodiazepine treatment. The benzodiazepine class of drugs also interacts with peripheral benzodiazepine receptors. Peripheral benzodiazepine receptors are present in peripheral nervous system tissues, glial cells, and to a lesser extent the central nervous system. These peripheral receptors are not structurally related or coupled to GABAA receptors. They modulate the immune system and are involved in the body's response to injury. Benzodiazepines also function as weak adenosine reuptake inhibitors. It has been suggested that some of their anticonvulsant, anxiolytic, and muscle relaxant effects may be in part mediated by this action. Benzodiazepines have binding sites in the periphery, however their effects on muscle tone is not mediated through these peripheral receptors. The peripheral binding sites for benzodiazepines are present in immune cells and the gastrointestinal tract.

When a compound has limited solubility in water it is common practice (in the pharmaceutical industry, for example) to determine pKa values in a solvent mixture such as water/dioxane or water/methanol, in which the compound is more soluble. In the example shown at the right, the pKa value rises steeply with increasing percentage of dioxane as the dielectric constant of the mixture is decreasing. A pKa value obtained in a mixed solvent cannot be used directly for aqueous solutions. The reason for this is that when the solvent is in its standard state its activity is defined as one. For example, the standard state of water:dioxane mixture with 9:1 mixing ratio is precisely that solvent mixture, with no added solutes. To obtain the pKa value for use with aqueous solutions it has to be extrapolated to zero co-solvent concentration from values obtained from various co-solvent mixtures. These facts are obscured by the omission of the solvent from the expression that is normally used to define pKa, but pKa values obtained in a given mixed solvent can be compared to each other, giving relative acid strengths. The same is true of pKa values obtained in a particular non-aqueous solvent such a DMSO. A universal, solvent-independent, scale for acid dissociation constants has not been developed, since there is no known way to compare the standard states of two different solvents.
=== Natural fission chain-reactors on Earth === Criticality in nature is uncommon. At three ore deposits at Oklo in Gabon, sixteen sites (the so-called Oklo Fossil Reactors) have been discovered at which self-sustaining nuclear fission took place approximately 2 billion years ago. French physicist Francis Perrin discovered the Oklo Fossil Reactors in 1972, but it was postulated by Paul Kuroda in 1956. Large-scale natural uranium fission chain reactions, moderated by normal water, had occurred far in the past and would not be possible now. This ancient process was able to use normal water as a moderator only because 2 billion years before the present, natural uranium was richer in the shorter-lived fissile isotope 235U (about 3%), than natural uranium available today (which is only 0.7%, and must be enriched to 3% to be usable in light-water reactors).
In 2010 ISB partnered with the Ohio State University Wexner Medical Center in Columbus, Ohio, to establish the nonprofit P4 Medicine Institute (P4MI). Its goal was stated as being "to lead the transformation of healthcare from a reactive system to one that predicts and prevents disease, tailors diagnosis and therapy to the individual consumer and engages patients in the active pursuit of a quantified understanding of wellness; i.e. one that is predictive, preventive, personalized and participatory (P4)." In 2012, P4 Medical Institute established an agreement with its first community health partner, PeaceHealth. PeaceHealth is a not-for-profit Catholic health care system, operating in a variety of communities in Alaska, Washington and Oregon. In 2016, ISB affiliated with Providence Health & Services, and Hood became the senior vice president of Providence St. Joseph Health and its chief science officer. Hood has published more than 700 peer-reviewed papers, received 36 patents, and co-authored textbooks in biochemistry, immunology, molecular biology, and genetics. In addition, he co-authored, with Daniel J. Kevles, The Code of Codes, a popular book on the sequencing of the human genome. He has been instrumental in founding 15 biotechnology companies, including Amgen, Applied Biosystems, Systemix, Darwin, Rosetta Inpharmatics, Integrated Diagnostics, and Accelerator Corporation. In 2015, he co-founded a startup called Arivale offering a subscription-based 'scientific wellness' service which shut down in 2019.
Sambunigrin, also a glycoside of mandelonitrile, is found in several species of the genus elderberry (Sambucus), including black elderberry and Canadian elderberry, as well as in Ximenia americana. Vicianin, another mandelonitrile glycoside, occurs in ferns of the genus Davellia (family Davalliaceae). Dhurrin is a cyanogenic glycoside of 4-hydroxymandelonitrile found in sorghum millet and other species of the genus sorghum millet, including Sorghum halepense. Linamarin (with the aglycone acetone cyanohydrin) and lotaustralin (with the aglycone butanone cyanohydrin) occur in the genera Linum (for example in common flax) and lotus flowers, as well as in the common bean. Both compounds are also present in cassava. The mistletoe species Loranthus micranthus (genus Loranthus) contains linamarin gallate, a derivative in which linamarin is additionally esterified with gallic acid. The rubber tree also contains linamarin; studies indicate that in this case the compound likely serves as an important storage substance in addition to its defensive function. The seeds contain particularly high concentrations, and during seedling development the compound is metabolized without releasing hydrocyanic acid, suggesting utilization in other biosynthetic pathways.
== Diagnosis == Substance use disorders can be confused with other psychiatric disorders. There are diagnoses for substance-induced mood disorders and substance-induced anxiety disorders and thus such overlap can be complicated. For this reason, the DSM-IV advises that diagnoses of primary psychiatric disorders not be made in the absence of sobriety (of a duration sufficient to allow for any substance-induced post-acute-withdrawal symptoms to dissipate) up to 1 year.

The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
Semax is a synthetic analog of the ACTH(4-10) fragment rather than a copy of the full hormone. Truncation plus the added tail removes the steroidogenic portion of ACTH. That distinction explains why it is studied for nervous-system effects instead of adrenal stimulation.
It is registered as a prescription product in Russia and has been used there clinically, chiefly by the intranasal route. No approval exists in the United States or the European Union. Availability elsewhere typically falls under research-chemical distribution, with varying purity documentation.
Dry powder is usually kept at about -20 degrees Celsius for long-term storage and at 2 to 8 degrees Celsius for short working periods. Containers should stay sealed and protected from light and moisture to limit degradation.
